I'm search for a backpack that I can bring to work, but also carry enough clothes in for weekend trips. Ideally, I'd like something with a separate laptop compartment. Black or blue preferred, but not required.

I own this backpack, and it's got everything you're looking for. The dedicated laptop section has its own zipper, so you can grab your laptop easily. The main compartment is huge, and it zips open really wide, so it works well. It's also fairly waterproof, which is nice.

If you're willing to go with a roll-top bag, the Spire is fantastic. It's got a laptop compartment that fits 15" laptops and enough space for a few clothes, though you probably couldn't fit those and an extra pair of shoes.

I own this bag
I really love my Thule backpack. I have the 34 L bag, but there are other sizes as well. It's got a laptop section like you need and plenty of room for a weekend's worth of clothes.

I bike to work with this bag every day
Biking to work means having to deal with the occasional rain storm. I wanted a water resistant bag so I knew my laptop wouldn't get soaked when I did. I liked that this bag was top loading but also had a separate compartment for the laptop so I didn't need to take everything out of my bag just to get my computer out. The bag is big enough to carry my gym stuff along with my computer, but it's not bulky. The only downside is the Flex Vent on the back of the bag that is supposed to help cool your back and not be as hot doesn't help much, but that's pretty much the case with any commuting backpack. All in all, very happy with this purchase.
